Description:

Glutamate receptors constitute the principal excitatory neurotransmitter receptors in brain. Two classes of glutamate receptors exist: Ionotropic receptors, and metabotropic receptors (mGluRs). Ionotropic glutamate receptors are oligomeric complexes of various subunits (GluR1 7, NMDA1 3, KAI 2) which comprise ligand gated calcium channels.

Background

Function:

G-protein coupled receptor for glutamate. Ligand binding causes a conformation change that triggers signaling via guanine nucleotide-binding proteins (G proteins) and modulates the activity of down-stream effectors. Signaling activates a phosphatidylinositol-calcium second messenger system and generates a calcium-activated chloride current. Plays an important role in the regulation of synaptic plasticity and the modulation of the neural network activity.

Western blot analysis of GRM5 expression in HT-29 cells,The lane on the left was treated with the antigen-specific peptide.
AF5243 staining A-431 cells by ICC/IF. Cells were fixed with PFA and permeabilized in 0.1% saponin prior to blocking in 10% serum for 45 minutes at 37°C. The primary antibody was diluted 1/200 and incubated with the sample for 1 hour at 37°C. A Alexa Fluor 594 conjugated goat polyclonal to rabbit IgG (H+L), diluted 1/600 was used as secondary antibody.

Function :

Blocking peptides are peptides that bind specifically to the target antibody and block antibody binding. These peptide usually contains the epitope recognized by the antibody. This mechanism is useful when non-specific binding is an issue, for example, in Western blotting (immunoblot) and immunohistochemistry (IHC). By comparing the staining from the blocked antibody versus the antibody alone, one can see which staining is specific; Specific binding will be absent from the western blot or immunostaining performed with the neutralized antibody.
