Figure 3ABHD7 is the depalmitoylase of lamin A (A) Exogenous ABHD5, -7, -17A, -17C, and -18 interact with lamin A. Myc-tagged lamin A was co-expressed with FLAG-tagged ABHDs, PPTs, or APTs in HEK293T cells, followed by immunoprecipitation for western blotting. (B) ABHD7 interacts with endogenous lamin A. Endogenous lamin A was detected by western blotting in HEK293T cells expressing FLAG-tagged ABHDs. (C) Overexpression of ABHD7 reduces palmitoylation of lamin A in HEK293T cells. The palmitoylation of lamin A was detected by ABE assay in HEK293T cells expressing FLAG-tagged lamin A and Myc-tagged ABHD7. “Ratio,” biotin/FLAG. (D) Knocking down ABHD7 increases exogenous palmitoylation of lamin A. The palmitoylation of FLAG-tagged lamin A was detected in ABHD7-knockdown HEK293T cells through ABE assay. “Ratio,” biotin/FLAG.
